There are serious concerns about the use of viral vectors for in vivo gene transfer, especially gene therapy, such as oncogenic potential and high immunogenicity. Accordingly, an easy, safe, non-toxic and possibly efficient delivery means of genes to a specified target tissue has been an attractive issue for researchers. In this sense, in vivo electroporation (EP) is thought to be one of the promising approaches since : handling is simple, quick, i.e., within a matter of seconds, and easy; transfection efficiency is probably higher than any other nonviral method; immunogenicity is low; there may be no limitation in size and concentration of DNA transferred, and ; no special preparation of DNA is required. Although gene expression exerted is transient when transfected by in vivo EP, foreign gene expression may last as long as more than 1 year.
In EP procedures, tissues of living animals are subjected to electric field that causes not only membrane pores but also shape deformation of cells, leading to the induced membrane permeability, hence transport of large molecules such as DNA across the cell membrane. To my best knowledge, no extensive and systemic analyses of EP parameters have been conducted so far, and therefore, most experimental conditions have been empirically known. Nevertheless, the results obtained are implicative that in vivo gene EP could be applied to a wide variety of experimental and therapeutic purposes. In this brief talk, some examples are presented in relation to gene therapy, growth promotion, pharmaceutical protein production and embryo manipulation. |
