Sequencing TempliPhi product: TempliPhi uses bacteriophage
Phi29 DNA polymerase and rolling circle amplification to produce
microgram quantities of template DNA from picograms of input without
cultured growth. The TempliPhi reaction was performed offline of
the Parallab 350TM. Sequencing reaction conditions:
600nl reaction volume, 60ng template, 1.25pmol primer, 190nl Big
Dye, thermal cycling conditions: 96℃ for 20 sec then (96℃/0sec;
50℃/0sec; 60℃/45sec) X 45 cycles, SPRI purified; and sequenced on
an ABI 3730xl. PHRED 20 = 880 bases |
